Okay, after spending ages fixating without contributing, here's my take:

Antagonist: Dunno witch species but a self-proclaimed Demi-God/ess [of insert ancient empire] with not much to back up his/her villainy. As a result comes off comical.
"The Mumbo":It'd be cool if this character was a ringtailed lemur (perhaps wearing a skull), possibly with a dislike of Laylee. Given his species' nocturnal, it may justify the character napping when you enter his tree (found in over half the worlds, all worlds in Twooka). Not sure what about his tokens, maybe fruits with his face pattern/shape?
The "Jinjos": Different species of parrots?
Fast travel: Stumped on this but perhaps burrows?
The "Chuffy": As a nod to Project Dream it would be cool if you acquire a pirate ship after defeating the boss of a beach level. Consequently most worlds and over world would need to feature open bodies of water. In hindsight I guess I've been playing to much AC Black Flag, but hey!

Just thought of something.

Yooka should be able to use her tongue to propel himself over huge gaps. By grappling to certain object and then pulling back the controller... And then release! Just like a slingshot.

I think another cool thing is if you could unlock costumes for the gang once certian achievements are met. Like you could get a Indiana Jones-esce costume, and like a "cool kidz" hat for Yooka and sunglasses for laylee, which could also be used in the rap at the end. There could even be like an indian costume.

This could work also because of the multiplayer, since you could also dress up your yooka and laylee to how you want, so you can tell what it looks like.

This idea won't make the game, but it would work in this platformer (to make the logistic hell even grander)

Interchangable partners, the real 'Stop n Swop'

--- You stop
--- Savour the moment
--- Change Laylee out for a different partner

The game could feature more then one partner, with new moves! Marvelous right?

A spider able to shoot webstrings, for Yooka to walk or climb on/over.
the spider can be mounted, to cross new surfaces.

Trowser the snake, no idea what he could do

A fairy, that tells you to listen once every 20 seconds

The only reason I don't like the swap partners idea is because they've said Yooka and Laylee will be pretty equal stars of the show, as opposed to BK where Banjo was really only empowered by Kazooie.

Now building on that idea... Laylee transformations, BK-style. Think Dragon Kazooie. Laylee could become a spider or snake or rhino or whatever.

Something I just considered regarding the mumbling voice acting.

They could pull somewhat of a "Jay and Silent Bob" where they drop JUST ONE plain spoken English line into the game as a punchline somewhere.

I can't even come up with example dialogue but it would essentially be like the "no ticket" part from Dogma. The characters speak in jibberish all game, but ONE LINE somewhere is just blatant plain English. I think it could be pretty funny.

So playtonic said they were gonna do something special with the soundtrack. Me being super bias to david wise ( being my favorite composer of all time) thought , hey i wish he had the full soundtrack then i thought. WHAT IF you could change between david's and grant's soundtrack , as they would both make a full length soundtrack but you the player could pick what sountrack you wanna play with. This would also warrant different playthroughs . just an idea
